CARBOHYDRATE METABOLIC ENZYMES OF STREPTOZOTOCIN INDUCED DIABETES MELLITUS IN KIDNEY OF ALBINO WISTAR RATS TREATED WITH CALLUS AND LEAF EXTRACTS OF Pergularia daemia (FORSSK.) CHIOV

ABSTRACT The study was carried out to evaluate the carbohydrate metabolic enzymes of hexokinase, glucose-6-phosphatase and fructose-1, 6- phosphatase from streptozotocin induced diabetic animals treated with chloroform, ethanol, aqueous and ethanol leaf callus extract of Pergularia daemia. The animals are treated with various concentration of chloroform, ethanol and aqueous extract are in a dose dependent manner such as 100 mg/kg, 200 mg/kg and 300 mg/kg and glibenclamide 600 μg/kg for 21 days. The ethanol leaf callus extract had significantly higher metabolic effect than leaf ethanol, aqueous and chloroform extracts. The carbohydrate metabolic enzymes activity of hexokinase in chloroform leaf extract of P. daemia were 0.055 (μmol glucose phosphorylated/h/mg protein) and ethanol leaf extract 0.065 and aqueous leaf extract 0.062 and ethanol leaf callus extract were 0.086 and glibenclamide is 0.071. The glucose-6- phosphatase of various extracts were 0.239 (μmol of phosphorylated/min/mg protein), 0.174, 0.187, 0.161 and 0.205. The fructose-1, 6-phosphatase were 0.480 (μmol of phosphorous liberated/min/mg of protein), 0.406, 0.427, 0.395 and 0.502 respectively. There was no toxic changes were observed in leaf extract (100 mg/kg, 200 mg/kg and 300 mg/kg) treated rats alone. The results obtained in this study indicate that the Pergularia daemia leaf extract can be a potential source of carbohydrate metabolic enzymes in kidney.
