FORMULATION & CHARACTERIZATION OF CHLORZOXAZONE SOLID DISPERSIONS

ABSTRACT
Chlorzoxazone is a centrally acting muscle relaxant and it is poorly water soluble drug. This article investigates enhancement of the dissolution profile of Chlorzoxazone using solid dispersion (SD) with PEG4000 by using different methods. 1:1, 1:2, 1:3, 1:4 and 1:5 ratios of solid dispersions were prepared by using different method such as Physical mixture, Solvent evaporation & Fusion techniques. Dissolution studies using the USP paddle method were performed for prepared solid dispersions of Chlorzoxazone at 37±0.5oC and 50 rpm in Phosphate buffer of pH 8. Fourier transformer infrared (FTIR) spectroscopy and x-ray diffractometry (XRD) were performed to identify the physicochemical interaction between drug and carrier, hence its effect on dissolution. Dissolution of Chlorzoxazone improved significantly in solid dispersion ratio of 1:5. Dissolution studies of the
preparations revealed that solid dispersions made of drug polymer in the ratio of 1:5 has much amount of drug released at the end of 1 hour and Solvent evaporation method was selected as best method among the methods. Thus, the solid dispersion technique can be successfully used for improvement of dissolution of Chlorzoxazone.
