Abstract

A REVIEW ON CRYSTALLO-CO-AGGLOMERATION OF LOW DOSE DRUGS

ABSTRACT

The term direct compression was long used to identify the compression of a single crystalline compound into a compact without the addition of other substances. Ingredients should show good flow, cohesion, and lubricating properties under pressure to make such compacts possible. Some of the NSAIDs exhibit poor compressibility and flowability and were not suitable for direct compression. But low dose drugs under NSAID category may be best suitable agents for direct compression by applying crystallo co agglomeration technique. Crystallo co agglomeration (CCA) is an innovative technique developed with intention to provide the drugs with good micromeritic and mechanical characteristics. The process of crystallo co agglomeration involves crystallisation followed by simultaneous agglomeration of the drug with the aid of a good solvent and /or a bridging liquid and a bad solvent. This process enables designing of spherical agglomerates containing low dose, poorly flowable and compressible drugs and also reduces processing time and thus may reduce labor costs, manufacturing steps and pieces of equipment. Less process validation and a lower consumption of power are some of the additional benefits of crystallo co agglomeration process in direct compression.
