FORMULATION AND EVALUATION OF GASTRIC FLOATING DRUG DELIVERY SYSTEMS OF CARVEDILOL WITH NATURAL POLYMERS

ABSTRACT
The present investigation was concerned with the development of oral
gastroretentive floating tablets of carvedilol, which after oral
administration were expected to prolong the gastric residence time and
improves the bioavailability of the drug as well as its half life. All the
precompression parameters of formulation blends of Carvedilol
floating matrix tablets were evaluated for Angle of repose, Bulk
density, Tapped density, Compressibility index and Hausner’s ratio.
All the values were within the limit as per U.S.P. The best floating
performance and the best in vitro drug release profile were achieved by
formulation F4 which contains Drug: xanthan gum: karaya gum in a
ratio of 1:0.5:0.5 gave the drug release (99.64%) up to 24 h. For the
optimized formulation, based on R2 values the best formulation follows zero order kinetics
and Higuchi model and suggests the release occurs by diffusion mechanism, based on ” n”
value it follows non fickian diffusion mechanism. Therefore matrix tablets with sufficient
floating time and sustained release up to 24 hrs were successfully prepared.
