EVALUATION OF THE EFFECT OF POLARITY OF PLANT POLYPHENOLS AND REACTION CONDITIONS ON FORMATION OF SUPRAMOLECULAR LIPID COMPLEX BY SOPHISTICATED CHROMATOGRAPHIC METHOD

ABSTRACT
Objective: In this study, we aim to evaluate the effect of polarity of plant polyphenols like catechin, quercetin and myricetin on formation of lipid complex. Different temperature conditions were maintained during the reaction, to examine the effect of reaction conditions on formation of the complex. Method: two type of reaction conditions were maintained for formation of polyphenol-lipid complex. Reaction was carried out at different tempertures. In Method I 25°C and in Method II 4°C was maintained. A gradient HPLC method was used for monitoring the formation of supramolecular lipid complex. A reverse phase Qualisil BDS-C18 column (4.6mm φ x 25. mm) packed with 5μm diameter particles was used. Following mobile phases were used for the study- Phase- A = Acetonitrile Phase -B = Acetonitrile: water (10:90) buffered to maintain pH = 3.20 with O-phosphoric acid. Mass
spectra was also recorded for phospholipids, standard polyphenols and lipid complex. Results: HPLC chromatograms of supramolecular lipid complexes obtained by Method I and II were compared. When compared to Method II more quantitiy of catechin and myricetin were left unreacted in Method I. Conclusion: Quercetin being relatively non polar forms complex with lipid in both the conditions whereas for polar polyphenols like catechin and myricetin low temperature conditions need to maintained. Thus non-polar polyphenols form lipid complex easily compared to polar polyphenols.
