Abstract

FORMULATION, DEVELOPMENT AND EVALUATION OF SUSTAINED RELEASE BEADS OF PREGABALIN.

ABSTRACT

Objective: The present study is aimed to develop sustained release beads of Pregabalin using ionotropic gelation method by utilizing a hydrophobic polymer which will form hydrophobic matrices to get delayed drug release. Method: Different concentrations of Compritol ATO 888 and Sodium Alginate were utilized to achieve desired characteristics of optimized batch. A 32 full factorial design was employed for optimizing the concentration of hydrophobic polymer. In this design, concentration of Compritol ATO 888 (X1) and concentration of Sodium Alginate (X2) were selected as independent variables. All the batches were evaluated in terms of different parameters like entrapment efficiency, swelling index, % drug release, particle size, angle of repose, FTIR study and so on. Result: From all the batches, F5 batch was selected as the optimized batch using Design expert 10.0.5 with 81.25% Entrapment Efficiency, 22.35% drug release after 2 hours, 48.33% drug release after 8 hours, 71.14% drug release at 16 hours and 99.15% after the completion of 24 hours. The drug release followed non-fickion diffusion model. Conclusion: It can be concluded that the method used to formulate sustained release beads of Pregabalin is simple and acceptable. The developed formulation is stable and provides sustained release of the drug about 24 hours.
