FORMULATION AND EVALUATION OF IMMEDIATE RELEASE TABLETS OF GLIMEPIRIDE BY USING SOLID DISPERSION TECHNIQUE USING GELUCIRES

ABSTRACT
Glimepiride is one of the third generation sulfonylurea used for
treatment of type 2 diabetes. Poor aqueous solubility and slow
dissolution rate of the glimepiride lead to irreproducible clinical
response or therapeutic failure in some cases due to sub therapeutic
plasma drug levels. In this study solid dispersion of the drug were
prepared by melting method and the drug was taken with carrier in the
proportions of (1:3, 1:4 and 1:5). The rate of dissolution of Glimepiride
was increased with the proportion of (1:5) (Drug: Gelucire 55/18)
when compared to the other formulations. Solid dispersions of
Glimepiride with Different grades of Gelucire were prepared and
further compressed as tablets by using Croscarmellose sodium, Micro
crystalline cellulose pH 112, Lactose monohydrate, Hydroxy propyl
cellulose, Sodium steryl fumarate and Magnesium stearate. The immediate release tablets
were prepared by using SD_3 formulation. F5 is the better formulation when compared to all
formulations. The F5 formulation has given 95% drug release in 30mins and it is following
the first order kinetics.
