ABSTRACT

The purpose of this research work was to develop and evaluate transdermal patches containing antihypertensive drug Amlodipine besylate. Preformulation studies were carried out on the drug. Solvent casting technique was used to prepare the matrix diffusion controlled system. Nine formulations were prepared with 10mg of Amlodipine besylate with different ratios of hydroxyl propyl methyl cellulose and sodium alginate polymeric systems by using poly ethylene glycol as plasticizer. Physicochemical incompatibility of drug and polymers were studied by FT-IR spectroscopy which suggested the absence of any incompatibility. Formulated transdermal patches were evaluated with regard to thickness, weight variation, percentage flatness, folding endurance, percentage moisture loss, percentage moisture uptake, water vapour transmission rate, drug content and in-vitro release studies. In-vitro permeation studies of formulations were performed by using modified Franz Diffusion cell. All prepared formulations were of good physical stability. From all the studies performed it was found that formulation F7 shows optimum release.

Keywords: Amlodipine besylate, Solvent casting, Hypertension, Hydroxy propyl methyl cellulose, Sodium alginate, Polyethylene glycol.
