Abstract

FORMULATION AND IN-VITRO EVALUATION OF GASTRORETENTIVE FLOATING EFFERVESCENT TABLETS OF OFLOXACIN

ABSTRACT

This study aimed to develop and evaluate hydrophilic matrix based controlled release gastroretentive drug delivery system of ofloxacin. Effervescent floating gastroretentive and immediate release minitablets (IRMT) were manufactured by direct compression using microcrystalline cellulose as a superdisintegrant and sodium bicarbonate as an effervescent substance, each mini-tablet containing approximately 25 mg Ofloxacin. Sustained release mini-tablets (SRMT) were formulated using various ratios of HPMC K15M and Xanthum Gum. Capsules were filled with both immediate release minitablets and sustained release mini-tablets to deliver 100 mg of Ofloxacin. Minitablets containing Ofloxacin IRMT were shown to release nearly 98% of the drug within 60 minutes. Minitablets containing SRMT released 94% of Ofloxacin over 12 hours. The feasibility of delivering a drug in biphasic immediate and sustained release manner was reputable by combining floating mini-tablets in a single hard gelatin capsule. The minitablets were evaluated for parameters such as Weight variation, Hardness, Friability, Drug content, Swelling index, in vitro buoyancy study and in vitro drug release study. The method adopted for in vitro drug release studies was USP Method-II (Rotating Paddle Method) by Pharma test dissolution apparatus using phosphate buffer 6.8 pH as a dissolution medium. Based on the evaluation results, F7 formulations was found to be the best of formulations and was checked for stability as per ICH guidelines.
