Abstract

DESIGN AND IN VITRO CHARACTERIZATION OF NOVEL PHASE TRANSITION SYSTEMS FOR NASAL DRUG DELIVERY

ABSTRACT

The purpose of this study was the design and in vitro characterization of phase transition systems for nasal drug delivery based on muco-adhesive polymers. Muco-adhesive drug delivery systems that utilized the property of bioadhesion of certain polymers, which becomes adhesive on hydration and hence can used for targeting a drug to a particular region of the body for extended periods of time. Phase transition systems of Ropinirole HCL for nasal drug delivery were prepared with muco-adhesive polymer such as chitosan and various grades of Hydroxyl Propyl Methyl Cellulose (HPMC) sodium beta glycerophospate. Chitosan hydrochloride precipitates to form hydrated gel above pH 6.5. Thus temperature induced precipitation was attained by beta glycerophosphate, which keeps the chitosan in hydrated, solution form by getting incorporated between the chitosan molecules and avoiding hydrophobic interaction at lower temperature while dehydration at higher temperature leads to geletion. Phase Transition System was evaluated for different parameter and result of optimized batch such as %Drug Content(100.53±0.06), Gelation Temperature(35-370C), The gelation time(7-8min), Viscosity Measurements (21.4±0.43cp), Rheological Behavior (Pseu-doplastic flow), Duration of Mucoadhesion (6.85±0.21 Hours ), Mucoadhesive force(34.965± 0.63 dynes/cm2 x 103),In- Vitro Release Study(96.92±0.11 for 420min) and In-Vitro Permeation Study(90.01±3.13 for 420min), Stability Study.
