Abstract

FORMULATION AND EVALUATION OF SUSTAINED RELEASE VILDAGLIPTIN MATRIX TABLETS BY USING PECTIN NATURAL POLYMERS AS RELEASE RETARDANT

ABSTRACT

The purpose of this research work was to establish Vildagliptin sustained release matrix tablets of 50mg. Vildagliptin is an anti diabetic drug of the new dipeptidylpeptidase-4 (DPP-4) inhibitor class of drug. Vildagliptin is one of emerging short acting drug, so developed formulation provides the advantages of sustained release formulations. The natural polymers like pectin were utilized in the formulation of matrix tablets containing vildagliptin by wet granulation technique and evaluated for its drug release characteristics. Pectin is hydrophilic and rate controlling polymers. Granules were prepared and evaluated for Bulk density, tapped density, compressibility index and angle of repose, shows satisfactory results. Formulation was optimized on the basis of acceptable tablet properties hardness, friability, drug content, weight variations, FT-IR studies and in vitro drug release. All the formulations showed compliance with pharmacopieal standards. Six formulations of different drug: polymer concentrations were formulated. First three formulations contain drug with pectin polysaccharide and other three formulations contains drug with polymer combination of pectin: HPMC K100M -F1 (1:1), F2 (1:2), F3 (1:3) F4 (1:1), F5 (1:2) and F6 (1:3).Among all the formulations, F6 which shows better sustained release at the end of 12 hrs. it has been found that the optimized formulation F6 containing (pectin and HPMC K100M) shows better sustained effect for 12 hr when compared to other formulations. The results indicated that a decrease in release kinetics of the drug was observed by increasing the polymer concentration. The release data was fitted to various mathematical models such as, Higuchi, Korsmeyer-Peppas, First-order, and Zero order to evaluate the kinetics and mechanism of the drug release.The drug release of optimized formulation F6 follows zero order kinetics.
