ABSTRACT The present study deals with the estimation of serum creatinine by Jaffe’s alkaline picrate method. Creatinine is a waste product formed in muscle by creatine metabolism. Creatine and creatine phosphate are spontaneously converted to creatinine at a rate of about 2% the total per day. This method helps in determination of creatinine excreted by a reported UV-Visible Spectrophotometric method and Colorimetry(a modified UV-Visible Spectrophotometric) method. The present article describes that creatinine present in serum, in alkaline medium reacts with picric acid to form a red tautomer of creatinine picrate the intensity of which is measured at 496nm. Hence, the main objective of the study was to compare the reliability of the two detection methods factors like wavelength and conditions of colour development influencing while this reaction was examined. The results of colorimetry method showed that the factors- wavelength detection and trough of creatinine picrate were over-come and it proved to be a more reliable method. Calibration curves were plotted to check for the creatinine concentration in given serum samples.The colorimetric method for serum creatinine estimation proves to be a more trustworthy method on laboratory scale basis.
