Abstract

HYPOGLYCAEMIC EFFECT AND INTERACTIONS OF ANGIOTENSIN II TYPE I RECEPTOR BLOCKER (TELMISARTAN) WITH ORAL HYPOGLYCAEMIC AGENTS IN STREPTOZOCININDUCED DIABETIC RATS

ABSTRACT

Objective: To study the effect of angiotensin II receptor blocker on blood glucose level and possible interactions when combined with Oral hypoglycaemics in Streptozocin induced diabetic rats. Material & Methods: Present study performed in animal house of department of pharmacology G.S.V.M. medical college Kanpur with prior permission from Institutoinal animal ethics committee. Rats were divided into 9 groups & each group contains six rats. Diabetes was induced by a single intra peritoneal injection of streptozocin (STZ) at a dose of 60 mg/kg body weight dissolved in 0.01 M citrate buffer.Three days after STZ injection rats with blood sugar level more than 250 mg/dl were selected. Results: On comparison to diabetic control group (table 2) it was found that gliclazide exhibit significant hypoglycemia (P<0.05) throughout the period of observation, Metformin exhibit significant hypoglycemia (P<0.05) at 1 and 2 hrs, pioglitazone exhibit highly significant hypoglycemia (p<0.01) throughout the period of observation, Effect of Telmisartan on blood glucose level is significant (p<0.05) throughout the period of observation, table 9 and fig. 2, observations indicated that Telmisartan enhances the hypoglycemic effect of gliclazide significantly (P < 0.05).Table 11 and fig. 3, observations indicated that telmisartan enhances the hypoglycemic effect of metformin significantly (P < 0.05). Conclusion: From this study it is evident that ARBs may reduce the blood glucose level in normal and diabetic conditions. It was also observed that ARBs may also induce antihyperglycemic effect of oral antidiabetic drugs in experimental animal. It is therefore advisable to prescribe and use of ARBs with anti diabetic drugs carefully with necessary dose adjustment of doses to avoid adverse hypoglycemic episodes in diabetic individuals.
