ORODISPERSIBLE TABLETS: A COMPREHENSIVE REVIEW

ABSTRACT Orodispersible tablets (ODTs) have emerged as an innovative oral drug delivery system designed to overcome the swallowing difficulties associated with conventional tablets and capsules. These dosage forms rapidly disintegrate in the oral cavity upon contact with saliva, eliminating the need for water and providing greater convenience during administration. ODTs are particularly beneficial for pediatric, geriatric, dysphagic, bedridden, and psychiatric patients, thereby improving medication adherence and overall patient compliance. The formulation of ODTs requires careful selection of active pharmaceutical ingredients, superdisintegrants, excipients, taste-masking techniques, and manufacturing processes to achieve rapid disintegration while maintaining adequate mechanical strength and product stability. Various manufacturing approaches, including direct compression, freeze-drying (lyophilization), moulding, spray drying, sublimation, and three-dimensional (3D) printing, have been successfully employed to develop ODTs with improved quality and performance. This review discusses the fundamental aspects of ODT technology, including formulation principles, ideal characteristics, excipients, manufacturing methods, evaluation parameters, marketed products, recent technological advances, current challenges, and future perspectives. The review also highlights the growing importance of ODTs as a patient-friendly dosage form and their expanding role in enhancing the effectiveness of modern oral drug delivery systems.
