Abstract

A REVIEW FORMULATION AND EVALUATION OF CALCIUM CHANNEL BLOCKER DRUGS

ABSTRACT

Calcium channel blockers (CCBs) are among the most widely prescribed cardiovascular drugs for the management of hypertension, angina pectoris, cardiac arrhythmias, and other related disorders. Their therapeutic action is based on the inhibition of L-type calcium channels, which reduces calcium influx into vascular smooth muscle and cardiac cells, resulting in vasodilation, decreased peripheral vascular resistance, and improved cardiac function. This review provides a comprehensive overview of the formulation and evaluation of calcium channel blocker drugs, covering their mechanism of action, classification, pharmacokinetic properties, contraindications, adverse effects, drug interactions, and therapeutic applications. Particular emphasis has been given to formulation strategies aimed at improving the bioavailability, stability, and patient compliance of commonly used agentssuch as amlodipine. Various advanced drug delivery systems, including fast-dissolving tablets, floating drug delivery systems, and smart nanocarrier-based formulations, have been discussed for their ability to enhance drug release and therapeutic performance. In addition, the review summarizes evaluation parameters and findings from clinical studies that demonstrate the efficacy and safety of calcium channel blockers in cardiovascular therapy. Overall, recent advances in pharmaceutical formulation and drug delivery technologies continue to improve the clinical effectiveness of calcium channel blockers, making them an essential component in the long-term management of cardiovascular diseases.
