FLOATING DRUG DELIVERY SYSTEMS FOR GASTRIC REACTION

ABSTRACT Floating Drug Delivery Systems (FDDS) have gained considerable attention as an effective approach to improve the oral delivery of drugs that require prolonged gastric residence. Conventional oral dosage forms often pass through the gastrointestinal tract rapidly, limiting the absorption of drugs with a narrow absorption window or poor solubility at higher intestinal pH. Floating drug delivery systems overcome this limitation by remaining buoyant on gastric fluids for an extended period, allowing controlled and sustained drug release in the stomach. This review provides a comprehensive overview of FDDS, including their basic principles, mechanisms of buoyancy, classification, formulation approaches, evaluation parameters, advantages, limitations, and therapeutic applications. Various types of floating systems, such as effervescent, non-effervescent, raft-forming systems,floating tablets, capsules, microspheres, and alginate beads, have been discussed along with the role of polymers and gas-generating agents in maintaining buoyancy. Factors influencing gastric retention time and formulation performance are also highlighted. The review further emphasizes the importance of FDDS in enhancing bioavailability, reducing dosing frequency, improving patient compliance, and achieving site-specific drug delivery. Overall, floating drug delivery systems represent a promising and practical strategy for improving the effectiveness of oral drug therapy, particularly for drugs that benefit from prolonged gastric retention and controlled drug release.
