Abstract

FLOATING DRUG DELIVERY SYSTEMS: A REVIEW OF MARKETED PRODUCTS AND FUTURE DIRECTIONS

ABSTRACT

In the Pharmaceutical research the pharmaceutical industries are taking much interest in Gastro retentive drug delivery system that is Floating Drug Delivery System (FDDS) through the oral drug delivery system. The main aim of this study to review on FDDS focusing on its current advancement and its future. The Floating system works on the principle of buoyancy as they are low density system and have the ability to sufficiently buoyancy to flow over the gastric contents and remain buoyant in the stomach without affecting the gastric emptying rate for a prolonged period of time. Floating tablets and capsule are prepared by adding suitable ingredients with excipients like hydrocolloids, inert fatty materials and buoyancy increasing agents. Various categories of drugs like antacids, antidiabetic, antifungal and anticancer drugs are formulated into FDDS. FDDS have bulk density less than gastric fluids. Floating tablets have emerged as an effective approach for improving the oral delivery of drugs that require prolonged gastric residence time. These tablets are designed to remain buoyant in the stomach for extended period, allowing the drug to be released in a controlled manner while enhancing its absorption. This technology is useful for drugs that absorbed mainly in the upper GIT, have narrow absorption window and have poor bioavailability when administered orally. From past to recent years, considerable attention has been given to development of Floating drug delivery systems due to their potential to overcome the limitations of conventional oral preparations. Its formulation includes various techniques, including effervescent and non-effervescent systems that are used to achieve controlled drug release and buoyancy. There are several factors that affects the effectiveness of Floating tablets such as nature of drug, physiology of gastric fluid, condition of feeding, composition of formulation. Floating tablets has many challenges like gastric emptying and residence time but continue to be a promising strategy for gastroretentive drug delivery.
