COMPARATIVE STUDY OF FORMULATED SUSTAINED-RELEASE CAPSULES OF METFORMIN HYDROCHLORIDE WITH MARKETED FORMULATION USING CARBOPOL AND HYDROXYPROPYL METHYLCELLULOSE

ABSTRACT Sustained release drug delivery systems are developed to maintain consistent plasma drug concentrations and enhance patient compliance in the management of chronic diseases such as type 2 diabetes mellitus. The present study focuses on the formulation and comparative evaluation of sustained release capsules of Metformin hydrochloride using a combination of Carbopol and Hydroxypropyl methylcellulose as release-retarding polymers. Capsules were prepared by employing different polymer ratios to achieve prolonged drug release. The prepared formulations were evaluated for preformulation parameters such as flow properties and compatibility, followed by post-formulation studies including weight variation, drug content uniformity, and in vitro dissolution testing. Drug release data were analyzed using various kinetic models to determine the mechanism of release. A comparative study was performed with a marketed sustained release formulation to assess the performance of the developed system. The optimized formulation demonstrated a controlled and extended drug release profile comparable to the marketed product, along with acceptable physicochemical characteristics. The findings suggest that the combination of Carbopol and Hydroxypropyl methylcellulose is effective in achieving sustained release of metformin hydrochloride in capsule dosage form.
