Abstract

BIOPHYSICAL GATEKEEPING: A CRITICAL QUALITY ATTRIBUTE (CQA) FRAMEWORK FOR LIPOSOMAL DELIVERY SYSTEMS

ABSTRACT

Liposomal encapsulation has redefined the delivery of drugs and nutrients by mimicking natural biological membranes. Despite their potential, transitioning these microscopic vesicles from the laboratory to large-scale industrial production is often hindered by physical inconsistency. This review introduces a structured framework termed "Biophysical Gatekeeping," a protocol that replaces traditional methods with precise "formulation engineering." This approach ensures that every batch meets specific safety and performance standards before moving toward clinical use. Central to this framework is WBCIL’s LipoEdge™ technology, an advanced manufacturing platform designed to solve the industrial challenge of vesicle heterogeneity. LipoEdge™ ensures that Critical Quality Attributes (CQAs) are mathematically designed into the liposomal architecture. The process is organized into four sequential "Gates." First, Vesicle Architecture ensures particles are the correct size and shape. Second, Surface Chemistry validates the electrical charge and protective coatings to prevent clumping in the blood. Third, Compositional Efficiency confirms that at least 80% of the active ingredient is successfully encapsulated. Finally, Biological Validation tests survival through stomach acid and entry into the bloodstream. By following these gates, LipoEdge™ technology reliably bypasses the liver’s "first-pass" metabolism, significantly increasing the amount of nutraceuticals/pharmaceuticals that reach the body. This systematic protocol ensures that liposomes are not just simple transport vehicles, but robust, engineered guardians that protect their therapeutic payload until it reaches its destination.
