ANALYSIS OF RIZATRIPTAN BENZOATE AND MELOXICAM IN MIXTURE BY VALIDATED FIRST ORDER DERIVATIVE SPECTROPHOTOMETRIC METHOD
Priyanshi V. Darji*, Satish A. Patel
ABSTRACT A unique and precise first-order derivative ultraviolet spectrophotometric method was developed for the simultaneous estimation of Rizatriptan Benzoate and Meloxicam, either alone or in combination, has been developed using first order derivative UV spectrophotometry. Methanol was used as the solvent for the analysis in this study. Zero-order spectral analysis showed that Rizatriptan Benzoate and Meloxicam had their highest absorption at 225 nm and 360 nm, respectively. The zero-crossing technique transforms data into first-order derivative spectra to achieve selective quantification and overcome overlapping spectral data. The selected analytical wavelengths were 226 nm for meloxicam, corresponding to the zero-crossing point of Rizatriptan Benzoate, and 234 nm for rizatriptan benzoate, corresponding to the zero-crossing point of Meloxicam. The study found a direct link between concentration and the results, with both compounds yielding consistent results at concentrations between 2 and 12 μg/mL.The ICH Q2 (R2) guidelines require testing to assess linearity, accuracy, and precision through repeatability and intraday and interday testing while determining the limits of detection and quantification. The results achieved high accuracy because they remained within the expected range, while the %RSD values remained less than 2%. The recovery studies confirmed the reliability of the method through the successful demonstration of proper recovery rates.
