ASHWAGANDHA: AN EXTENSIVE REVIEW OF ASHWAGANDHA’S CHEMICAL CONSTITUENTS, BIOLOGICAL ACTIVITIES, AND THERAPEUTIC APPLICATIONS

ABSTRACT Winter Cherry is a widely used therapeutic plant utilized in ancient Ayurvedic practices, valued for its rejuvenating, adaptogenic, and restorative properties. This review provides a comprehensive overview of its phytochemistry, pharmacological actions, therapeutic applications, safety, and regulatory considerations. The plant is abundant in bioactive compounds including nitrogenous compound withanolide-type compounds, sitoindoside constituents, and various steroidal lactones, many of which contribute to its potent biological activities. Pharmacological studies demonstrate its inflammation-reducing, adaptogenic, free-radical-scavenging, immune-regulating, protective towards nerve cells, anticancer, stress reliving, and endocrine-regulating effects. Evidence from animal experiments and human clinical trials indicates its possible usefulness in managing health issues including stress abnormalities, neurodegenerative pathological conditions, infertility, hypothyroidism, inflammatory diseases, metabolic dysfunction, and cognitive impairment. Toxicological evaluations reveal a broad safety margin and low acute toxicity across various extracts. Despite growing scientific support, challenges persist regarding standardization, variability in bioactive constituents, dosage optimization, and regulatory harmonization. Future research should focus on identifying precise mechanisms of action, establishing robust quality-control frameworks, and conducting large-scale clinical trials to validate its therapeutic value. Overall, Ashwagandha emerges as a promising medicinal plant with significant potential for modern clinical and pharmaceutical applications.
