FORMULATION AND EVOLUTION OF SUSTAIN RELEASE DRUG DELIVERY SYSTEM OF TABLET

ABSTRACT Sustained release drug delivery systems (SRDDS) have emerged as a significant advancement in pharmaceutical technology, designed to deliver active pharmaceutical ingredients gradually over a defined period. By maintaining consistent drug concentrations within the therapeutic range, these systems reduce dosing frequency and improve patient adherence to treatment regimens. This review examines the core design concepts, types, and mechanisms of SRDDS, such as dissolution-controlled, diffusion-controlled, ion-exchange resin-based, pH-sensitive, and osmotic ally driven systems. It also delves into essential pharmacokinetic and pharmacodynamic factors—including half-life, absorption, metabolism, and distribution—that influence the development of such formulations. The discussion extends to both the benefits and limitations of sustained release systems, emphasizing their potential to enhance therapeutic outcomes and minimize adverseeffects. In addition, the pharmacological profiles, therapeutic applications, mechanisms of action, and potential side effects of several widely used SRDDS-compatible drugs—namely Aceclofenac, Diclofenac, Nitroglycerin, Metformin Hydrochloride, Etodolac, and Indapamide—are explored.
