ABSTRACT Advanced lipid-based drug delivery systems known as nanocochleates are created when negatively charged phospholipids and multivalent cations interact to form solid, spiral-shaped structures Unlike liposomes, which have an internal aqueous phase, nanocochleates provide a unique multilayered lipid matrix that protects encapsulated bioactive substances from enzymatic degradation and adverse environmental condition. Proteins, peptides, antivirals, anticancer medications, vaccines, and nutraceuticals are just a few of the many therapeutic molecules whose oral bioavailability and stability it improves. Nanocochleates with high entrapment efficiency and regulated release profiles can be prepared using a variety of techniques, including the hydrogel method, trapping technique, and dialysis approaches. Nanocochleates exhibit promise uses in the treatment ofdiseases like cancer, diabetes, malaria, and leishmaniasis as well as the delivery of nutraceuticals and vaccinations, despite obstacles like cost and storage needs.
