HEPATOPROTECTIVE PROPERTIES OF METHANOL EXTRACT OF DIALIUM GUINEESE LEAVES AGAINST CCL4-INDUCED HEPATOTOXICITY IN WISTAR RATS

ABSTRACT Liver diseases pose a significant burden on global public health, with diverse etiological factors contributing to hepatic injury and dysfunction. This study investigates the hepatoprotective properties of the methanol extract of Dialium guineese against CCl4 induced hepatotoxicity in wistar rats. This study was conducted for a period of 14 days using forty-eight (48) Wistar rats divided into 6 groups of 8 rats each. This grouping includes Group 1 (Normal control), Group 2 (Negative control received only CCl4 2.5 ml/kg b.w on day 13 and 14), Group 3 (Standard control), Group 4 to 6 were induced with CCl4 and treated with 100, 200 and 400 mg/kg bw of methanol extracts of D. guineense respectively. Glutathione and catalase levels exhibited a significant decrease in the untreated group compared to the normal control group. Conversely, treatment with methanol extracts of D. guineense leaf resulted in a significant increase in glutathione and catalase levels, albeit in a non-dose-dependent manner. Albumin levels demonstrated a significant increase in the untreated group compared to the normal control group. However, treatment with methanol extracts of D. guineense leaf resulted in a significant decrease in albumin levels. The electrolyte levels, chloride ions exhibited a significant increase in the untreated group compared to the normal control group. The assessment of liver function biomarkers revealed significant changes in enzyme and lipid peroxidation levels. The induction with CCl4 led to a significant increase in AST, ALT and ALP levels indicating hepatotoxicity. However, treatment with D. guineense extract and the standard drug silymarinz exhibited a significant reduction. The methanol extract of D. guineense leaves exhibits significant hepatoprotective and antioxidant properties. These results support the potential use of D. guineense as a natural therapeutic agent for the prevention and treatment of liver diseases induced by toxic agents like CCl4.
