ABSTRACT This review explores the advancements in 3D bioprinting of collagen-based constructs for tissue regeneration, highlighting its significance in regenerative medicine. Tissue engineering aims to restore damaged tissues through the creation of biological substitutes, utilizing biomaterials, cells, and signalling molecules. 3D bioprinting enables precise layer-by-layer positioning of biological materials, enhancing control over microarchitecture and cell placement, which is crucial for functional tissue regeneration. Collagen, as a predominant biomaterial, plays a vital role in maintaining tissue structure and facilitating cellular activities. However, its use as a standalone bioink is limited by challenges such as low viscosity and mechanical strength. Recent research focuses on developing composite bioinks that incorporate other materials to enhance performance while maintaining biocompatibility. Advances in bioprinting technologies, including extrusion-based, inkjet, and laser-assisted methods, have enabled the fabrication of complex tissue constructs. Despite the potential of collagen-based bioprinting in engineering functional tissues like skin, cartilage, and bone, challenges remain in printability, mechanical robustness, and regulatory compliance. Future perspectives include the integration of recombinant collagen, AI-driven bioprinting, and personalized medicine approaches, aiming to create dynamic, patient-specific tissue constructs. Overall, collagen-based 3D bioprinting presents a promising platform for scalable and clinically viable tissue-engineered therapies.
