Abstract

STUDY ON THE ANTIOXIDANT EFFECTS OF PICEATANNOL IN DIABETIC RATS WITH NONALCOHOLIC FATTY LIVER DISEASE

ABSTRACT

Objective: To investigate the effects of piceatannol (PIC) on the activity of glutathione peroxidase (GSH-Px) and the content of malondialdehyde (MDA) in diabetic rats with nonalcoholic fatty liver disease (NAFLD), as well as its impact on the protein expression of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and interleukin-6 (IL-6) in liver tissues. Methods: NAFLD rats model was established using a high-sugar, high-fat diet combined with intraperitoneal injection of streptozotocin (STZ). The successfully modeled rats were randomly divided into a model group (M group), a low-dose PIC group (PIC-L group), and a high-dose PIC group (PIC-H grou). Normal rats were used as the control group (NC group), with 10 rats in each group. The PIC-L and PIC-H groups were administeredPIC by gavage at 50 mg/kg and 100 mg/kg, respectively, while the NC and M groups were given an equivalent volume of saline, once daily for 4 weeks. Blood glucose (FBG) levels were measured. The levels of TNF-α, IL-1β, and IL-6 in the serum of NAFLD rats were determined using ELISA. The activity of GSH-Px and the content of MDA were measured using colorimetric and visible light methods. Results: Compared with the NC group, the M group showed significantly increased FBG levels (P < 0.05), significantly decreased GSH-Px activity in liver tissue homogenates, significantly increased MDA levels (P < 0.05), and significantly elevated serum levels of IL-1β, TNF-α, and IL-6 (P < 0.05). Compared with the M group, both the PIC-L and PIC-H groups exhibited reduced FBG levels (P < 0.05). The PIC-H group showed significantly increased GSH-Px activity (P < 0.05), significantly decreased MDA levels (P < 0.05), and significantly reduced serum levels of IL-1β, TNF-α, and IL-6 (P < 0.05). Conclusion: PIC can reduce FBG levels, alleviate inflammatory responses, and enhance antioxidant capacity in NAFLD rats, thereby exerting a protective effect on the liver in NAFLD rats.
