SIMULTANEOUS HYDROPHILIC INTERACTION THIN LAYER CHROMATOGRAPHIC (HITLC) METHOD FOR DETERMINATION OF AMLODIPINE AND FUROSEMIDE BINARY MIXTURE

ABSTRACT In this research a simple, rapid, selective and a new hydrophilic interaction thin layer chromatographic (HITLC) method has been developed for simultaneous determination of amlodipine and furosemide using (acetonitrile:methanol: acetic acid, 30:20:2.5 v/v/v) as a mobile phase and silica gel F254 plates as stationary phase. The factors that play important role in this mechanism were defined. Mostly effective factors include mobile phase compositions, mobile phase compositions ratio, saturation time, the lipophilicity of the investigated drugs and the presence of acetic acid. Rf value of amlodipine was 0.24 ± 0.02 while of furosemide was 0.84 ± 0.01. The correlation coefficients between the concentrations of the studied drugs and the peak area were 0.997 and 0.995 in the range of (100–1000 μg mL−1) for amlodipine and furosemide respectively. Limits of detection LOD were 30.83 μg mL−1 and 29.45 μg mL−1 while limits of quantitation (LOQ) were 92.49 and 88.35μgmL-1 for amlodipine and furosemide, respectively. The proposed method was validatedaccording to USP31 and used for the determination of the cited drugs in their binary mixture as pure or as co-administered tablets successfully.
