STUDY ON THE HEPATOPROTECTIVE EFFECT OF PICEATANNOL IN DIABETIC RATS BASED ON THE NF-?B PATHWAY
Song Wei, Liang Minghui and Sreemoy Kanti Das*
ABSTRACT Objective: To investigate the effects of piceatannol (PIC) on liver function and nuclear transcription factor (NF-κB) expression in diabetic rats, providing a basis for future research on the hepatoprotective effects of PIC in diabetic rats through the NF-κB pathway. Methods: Thirty-nine Wistar rats were adaptively fed for 1 week and randomly divided into a normal control group, a model group, and a PIC group, with 10 rats in each group. After successful modeling, the PIC group was administered PIC by gavage for 4 weeks, once daily, while the normal control group and the model group were given an equal volume of saline by gavage. After 4 weeks of administration, blood and liver tissues were collected from each group.Fasting blood glucose (FPG) levels were measured using the glucose oxidase method; serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), and alkaline phosphatase (AKP) were determined by enzyme-linked immunosorbent assay (ELISA); and NF-κB expression levels in liver tissues were detected by RT-PCR and compared. Results: After 4 weeks of administration, the model group showed the highest levels of FPG, ALT, AST, AKP, and NF-κB, followed by the PIC group, while the normal control group had the lowest levels. The differences among the three groups were statistically significant (P < 0.05). Conclusion: PIC treatment for diabetes can reduce blood glucose levels and improve liver function in diabetic rats. The hepatoprotective effect of PIC may be mediated through the regulation of the NF-κB signaling pathway. Keywords: Diabetes; Rat model; Piceatannol; Liver function; NF-?B signaling.
