FLOATING MICROSPHERES AS A NOVEL DRUG DELIVERY: A SYSTEMATIC REVIEW

ABSTRACT The gastro-retentive system is a promising oral drug delivery approach due to its ability to stay in the gastric region for an extended period. This system improves drug bioavailability by maintaining a consistent release rate, leading to more stable plasma drug concentrations. Among various strategies to achieve gastro-retentive properties, floating microspheres are the most widely used. These low-density systems possess buoyancy, allowing them to float on the gastric contents and remain in the stomach for prolonged periods. Floating microspheres are spherical, free-flowing powders with sizes ranging from 1 to 1000 micrometres. Their ability to float over the stomach contents ensures that the drug is released gradually, helping to maintain a sustained therapeutic effect. This controlled release can reduce fluctuations in plasma drug concentrations, enhancing the overall efficacy and reducing side effects. The review examinesdifferent types of microspheres and various preparation methods. The key advantage of gastro-retentive floating microspheres lies in their potential to provide prolonged gastric retention, which is crucial for drugs that require a longer absorption window in the stomach. This approach helps in improving drug release profiles and patient compliance. Thus, floating microspheres represent a valuable tool in the development of advanced oral drug delivery systems.
