ABSTRACT

One appealing approach to treating various illnesses is the repurposing of current medications or the investigation of their possible therapeutic value in novel indications. Finding novel ways to shorten the time needed for medication discovery is essential. Drug repurposing has become more significant in the modern era for finding novel therapeutic applications for pharmaceuticals that are already on the market. Repurposing is usually accomplished either systematically or serendipitously. However, the success rates of drug repurposing programs are not particularly high. We intended to discover critical factors contributing to the high attrition rate in such studies by meticulously examining certain failed repurposing attempts. Based on the core principles of knowledge and evaluation, we have identified four pillars to improve the success rate in medication repurposing initiatives, viz, good knowledge of the repurposed medicine’s pharmacological features and human-safe drugs to treat rare and difficult illnesses is innovative. It is a highly successful approach to finding and developing new drugs. Numerous pharmaceutical companies have repositioned outdated drugs to create innovative treatments. Compared to creating a new drug from scratch, repurposing existing medications is efficient, quick, economical, and typically has fewer risks. A significant advancement in drug development is drug repositioning or repurposing, which focuses on finding new applications for outdated therapeutic agents. More specifically, drug repositioning is the process of investigating new uses for a medication that has already received approval, whereas drug repurposing involves comprehensive redevelopment strategies based on the active drug's exact chemical structure as it was in the original product.

Keywords: Drug repurposing, serendipitously, old drugs, attrition, existing medications, new drug from scratch, meticulously, indications.
