FORMULATION DESIGN, IN-VITRO CHARACTERISATION OF TOLNAFTATE LOADED NAIL LACQUERS FOR THE MANAGEMENT OF ONYCHOMYCOSIS

ABSTRACT The most prevalent fungal nail infection that affects both finger and toe nails is onychomycosis. Onychomycosis can be effectively treated with antifungals. Oral antifungal drug delivery, however, is more hazardous and necessitates a longer course of treatment. It was out that medicated nail lacquers were less harmful and needed less time to work. In addition to treating nail infections in the fingers and toes, it also protects the nails. Therefore, creating nail lacquer for transungual administration of tolnaftate was the goal of the current study. Nail lacquer is a liquid solution that gives color, shine, and sparkle to the nails when applied. They are similar to nail polish but have higher viscosity and long-lasting effects. This research aims to formulate and evaluate an antifungal nail lacquer with Transungual drug delivery system for treatment onychomycosis induced by candida albicans. Since all other formulations can be easily removed off by rubbing or washing Lacquers are the most effective way to deliver drug transungual, by incorporating the transungual drug delivery system effective and systemic release of drug can be achieved. Different parameters were evaluated like organoleptic characteristics, solubility, drying time, non – volatile content, water resistance, drug content, smoothness to flow, in–vitro permeation studies and antimicrobial activities were studied. The compatibility studies were done using Fourier Transform Infrared Spectroscopy. Tolnaftate incorporated nail lacquer of five different concentrations were formulated using different excipients using a magnetic stirrer. The optimal formulation NL 5, was chosen based on the highest attractiveness. The formulation that was optimized demonstrated the best drying time, water resistance, flow smoothness, drug content, in-vitro permeation, and Compared to the control formulation, better antifungal efficacy against Candida albicans was noted. Accordingly, the study suggests that nail lacquer might be a superior substitute for transungual tolnaftate delivery.
