DETERMINATION OF GALLIC ACID CONTENT IN FOUR VARIETIES OF AMALAKI (PHYLLANTHUS EMBLICA LINN.) FRUIT BY HPTLC

ABSTRACT Background: Amalaki (Phyllanthus emblica Linn.) is an important medicinal fruit used since Vedic Kala. Gallic acid is a natural phenolic compound found in fruits of Amalaki. It is reported to have antimicrobial and anticancer activity and protects from gastrointestinal, cardiovascular, metabolic and neuropsychological diseases. Objective: To analyse content of gallic acid in four varieties of Amalaki fruit through quantitative HPTLC profile. Material & Methods: The quantitative determination of gallic acid was performed on TLC aluminium plates precoated with silica gel 60F-254 as the stationary phase. The linear ascending development was carried out in a glass chamber saturated with a mobile phase consisting of Dichloromethane: Ethyl acetate: Formic acid (5:4:1 V/V/V) at room temperature (25 ± 2°C). CAMAG TLC scanner 4 was used for Spectro densitometric scanning and analysis, in the absorbance mode, at 273 nm. Results: The linear regression analysis data for the calibration plots showed good linear relationship with r2 = 0.999 in the concentration range of 40-240 ng/spot with respect to the peak area.
