PHARMACOKINETICS OF METOPROLOL SUCCINATE SUSTAINED RELEASE TABLET PREPARED BY USING NOVEL DRUG RELEASE MODIFIERS

ABSTRACT Objective: To establish the pharmacokinetics of metoprolol succinate in the rabbit model for newly developed sustained release tablet prepared by using Eulophia herbacae and Remusatia vivipara tuber mucilage. Materials and Methods: Pharmacokinetic parameters of metoprolol succinate were investigated in rabbits (n=6). Three rabbits were used for testing prepared metoprolol succinate SR tablet (formulation R8) and another three rabbits for marketed SR tablet (PROLOMET-XL 100) for the study. Pharmacokinetic characteristics were estimated using plasma drug concentration time profiles. Results: The mean of maximum plasma drug concentration (Cmax), AUC and half-life (t1/2) was found to be 195.6 µg/ml, 3626.93 µg.h./ml, and 5.3 h respectively for formulation R8. The mean of maximum plasma drug concentration (Cmax), AUC and half-life (t1/2) was found to be 222.85 µg/ml, 4163.64 µg.h./ml, and 7.1 h respectively for PROLOMET XL 100. Conclusion: This indicates that formulation R8 is equivalent to that of the standard PROLOMET XL 100. Keywords: Metoprolol, Eulophia herbacae, Remusatia vivipara.
