TRIMETAZIDINE IMPROVES DEXAMETHASONE INDUC ED AORTIC ABNORMALITIES IN RATS

ABSTRACT Introduction:Insulin resistance (IR) is a critical factor in thepathogenesis and clinical progression of various metabolic disorders,including diabetes. This study seeks to investigate the impact oftrimetazidine (TMZ) on IR, as well as t o elucidate the potentialunderlying mechanisms involved. Methods: An in vivo IR model wasestablished using male Wistar rats, which were administered TMZ atdoses of 10 mg/kg and 20 mg/kg for a duration of 14 days.Subsequently, from day 7 to day 14 of th e study, the rats receivedintraperitoneal (i.p.) injections of dexamethasone (DEXA) at a dosageof 1 mg/kg. Results: The findings indicate a notable decline in aorticmalondialdehyde (MDA) level. Comparative analysis revealed that theaortic levels of nuc lear factor kappa B p65 subunit (NF κB p65) andinterleukin 6 (IL 6) were significantly diminished by TMZ whencontrasted with those treated with DEXA. Furthermore, TMZdemonstrated efficacy in mitigating aortic histopathological changesthrough the inhibition of oxidative stress related inflammation in rats.
