Abstract

PROTOCATECHUIC ACID AMELIORATES UUO-INDUCED RENAL INJURY IN MALE MICE VIA REDUCTION OF OXIDATIVE STRESS, INFLAMMATION AND FIBROSIS

ABSTRACT

Objectives: Chronic kidney disease (CKD) could result from obstructive nephropathy; a condition that arises from urinary flow obstruction in the kidney leading to renal inflammation and injury. Protocatechuic acid (PCA) is a natural phenolic compound that shows a promising protective effect in cardiovascular diseases. In the current study, the reno-protective effects of PCA were investigated in a mouse model of unilateral ureteral obstruction (UUO). Methods: Four groups of male C57BL/6J mice (10-12 weeks old) were used (n=6-8/group); Sham control, Sham/PCA (50 mg/kg I.P daily for two weeks), UUO and UUO/PCA. Results: PCA significantly improved the UUO-induced decline in creatinine clearance and excretion. Moreover, PCA attenuated renal TBARs, IL-6, matrix metalloproteinase-2 (MMP-2) and MMP-9 in UUO-induced renal injury. Furthermore, PCA also significantly attenuated UUO-induced increase in renal collagen IV and KIM-1 expression. Conclusion: These data suggest that PCA provides renal protection in murine model of UUO via reduction of renal oxidative stress, inflammation and fibrosis.
