Abstract

BIOACTIVITY POTENTIAL OF AQUATIC BACTERIAL METABOLITES AGAINST CLINICAL ISOLATES SOURCED FROM THE SOUTH-SOUTH ZONE OF NIGERIA ESTUARIES

ABSTRACT

Metabolites of microorganisms from deep waters may deliver the right solution in the face of rising antimicrobial resistance. The secondary metabolites sourced from the estuaries were evaluated using standard microbiological techniques. Extraction was done using the boiling method. The 16SrRNA region was amplified using the 27F and 1492R Primers. Sequencing was done using the BigDye Terminator Kit. Editing was done using Ridom Trace Edit version 9.0 and BLAST in the NCBI database. The organisms were subjected to nutrient broth fermentation medium and agitated for 120 hours and centrifuged at 4000rpm for 10 minutes. Solvent-solvent extraction was done and the extract concentrated. Providential rettgeri from Okrika, Vibrio fluvialis from Ibaka, and Vibrio furnissi from Brass were identified. Vibrio furnissi inhibited S. pyogenes, P. mirabilis, and P. aeruginosa at 72 hrs. At 96 hours, only S. aureus and P. mirabilis were inhibited. Vibrio fluvialis, at 96 hours inhibited S. aureus and S. pyogenes while at 120 hours, P. mirabilis and P. aeruginosa were inhibited. Providential rettgeri inhibited S. aureus, S. pyogenes, P. mirabilis and P. aeruginosa. At 120 hours, only P. mirabilis and P. aeruginosa were inhibited. Providential rettgeri had an MIC of 0.0625 and 0.25 against S. aureus and P. aeruginosa, respectively. Vibrio fluvialis had an MIC of 0.015625 and 0.0625 against S. aureus and P.aeruginosa. Vibrio furnissi had an MIC of 0.03125 and 0.125 against S. aureus and P. aeruginosa The secondary metabolites extracted from Providencia rettgeri, Vibrio fluvialis, and Vibrio furnissi demonstrated bioactivity against clinical isolates
