ABSTRACT

Background: Clotrimazole is an antifungal drug and having biological halflife of two hours, and have gastro- intestinal side effect while administered orally. The objective of this study to develop a transdermal drug delivery system for Clotrimazole with turmeric oil to make a target site specific drug delivery for fungal infection and reduce the Sensitization or irritation problem while using transdermal patch. Methods: Firstly, the turmeric oil is extracted from rhizomes of turmeric secondly, these transdermal patch were prepared by solvent casting method by using placebo method; using various combinations of polymer with solvent MDC and methanol in (1:1) ratio, polyethylene glycol 500 and propylene glycol is use as permeation enhancer and plasticizer. Compatibility study for all the prepared patch were analyzed, all the prepared patch were evaluated for weight variation, thickness, weight variation in moisture absorption, moisture uptake, folding endurance, flatness, drug content and in-vivo permeation study. Result: The formulation was uniform in their physical characteristics combinations of three polymers HPMC, EC, EUDRAGIT RS -100 implying excellent quality and uniformity in patch characteristics as compared to other combination of polymers, in the in vitro drug release studies for all formulation showed that in first dose of drug were released in 2 to 2.5 hrs. and complete dose were release (95%) were achieved 24hrs. Conclusion: Transdermal patch were successfully prepared of Clotrimazole and turmeric oil and their evaluation implied a good quality and uniformity in patch characteristics and these patch have potential aspects in therapeutic effect and offering advantages in reducing dose frequency for treatment, improving patient compliance and bio- availability.

Keywords: Clotrimazole, Turmeric oil, Eudragit RS 100, HPMC, EC, Transdermal drug delivery system.
