ABSTRACT

Introduction: To obtain accurate Minimum Inhibitory Concentration (MIC), Minimum Bactericidal Concentration (MBC) and Time Kill Curve (TKC) of Diploknema butyracea extract against Aggregatibacter actinomycetemcomitans(Aa), Porphyromonas gingivalis(Pg), and Fusobacterium nucleatum(Fn) of 0.2% Chlorhexidine mouthwash. Materials & methodology: MIC was carried out by using serial dilution method and culture of microorganisms were incubated with the same drug concentration to assess MBC. Time Kill Curve was analysed in an anaerobic chamber. Results: MIC of the methanolic extracts for pathogens ranged between 0.2 μl/ml and 100μl/ml. among the test pathogens Pg showed MIC of extract was 3.12 μl/ml, Fn was 12.5 μl/ml and Aa was 6.25 μl/ml. Among P. gingivalis showed lowest MIC for extract. MBC analysis Pg and Fn highly susceptible to leaf extract. The TKC assay indicated that, only Aa showed bacterial growth from 5 min to 2 hrs. Whereas 0.2% chlorhexidine as a control group showed no bacterial colony growth for Pg, Fn and Aa. Conclusion: Present analysis concluded that, Madhuca longifolia has antimicrobial properties against periodontal pathogens which can serve as an adjunct for prevention of periodontal disease. So further clinical trials on these leaves are required, which can be helpful in the definitive therapeutic uses of M. longifolia in treating periodontal diseases.

Keywords: Madhuca longifolia, Minimum Inhibitory Concentration, Minimum Bactericidal Concentration and Time Kill Curve, antimicrobial.
