ABSTRACT

Present study aimed to formulate bilayer Metformin HCl-Glimepiride extended release matrix tablet using different grades of hydrophilic polymers. Five batches were formulated using 500mg Metformin HCl and 1mg Glimepiride as once in a day bilayer extended release tablets which can provide effective drug release for 10-12 hours. Tablets were prepared by wet granulation for Metformin HCl and direct compression for Glimepiride. All the formulations were evaluated for pre-compression & post-compression parameters like thickness, hardness, friability, weight variation and In-vitro release study. Based on the results, it was found that formulation containing Methocel K100M and Blanose 7HF-PH in maximum quantity (F5) was identified as good and best formulation among all formulations developed for extended release tablets of Metformin HCl & Glimepiride. Batch No.F5 was found to obey first order drug release governed by diffusion & erosion of the matrix and showed very good stability profile as well.

Keywords: Metformin HCl, Glimepiride, bilayer, extended release tablet, hydrophilic polymers.
