Abstract

DESIGN OF PROGESTERONE IMPLANTABLE DRUG DELIVERY DEVICE FOR ESTRUS SYNCRONISATION USING GELATIN

ABSTRACT

In the present work, subdermal implants were prepared for the treatment of hormonal imbalance with progesterone. Progesterone was chosen as a model drug due to its wide spectrum of activity, low toxicity and high efficiency. Implants comprising plasticizer, glycerine and polymer Gelatin were formulated. The formulated implants were evaluated for drug content uniformity, thickness, weight variation, IR, microbial degradation studies. In-vitro release of implants was studies in PEG 400: Distilled Water (1:1) buffer pH 7.4 and stability studies was done at ambient temperatures for 3 months. In-vivo studies in animals were carriedout for polymer-tissue compatibility at subdermal region. The prepared Progesterone subdermal implants were found to contain uniform weight, thickness, drug content and in-vitro release of progesterone. Subdermal implants which were treated with formaldehyde for 24 hours were found to be sustaining drug release for a period of more than 8 days. In-vivo studies in sheep for polymer tissue compatibility showed encouraging results depicting no changes in tissue configuration histopathologically and compatible with the surrounding tissue of subdermal region.
