ABSTRACT

This research aimed to formulate gastro-retentive/floating matrix tablet of Dimenhydrinate by direct compression and melt granulation method using different polymers viz. HPMC K15M, HPMC K4M Sodium alginate, poloxamer 188, bees wax, ethyl cellulose, sodium bi carbonate etc. each having 50 mg drug strength. All the formulations were evaluated for pre-compression & post-compression parameters like thickness, hardness, friability, Swelling Ability, In vitro Buoyancy Studies, Adhesion Retention Period and In-vitro release study. Based on the results, it was found that formulation containing PVP K30, HPMC K15M, & Poloxamer 188 (D-6) was identified as good and better formulation among all formulations developed for floating tablets of Dimenhydrinate by direct compression method ; and formulations prepared using bees wax & HPMC K4M in combination (D-10) was identified as best formulation among all formulations developed for floating tablets of Dimenhydrinate by melt granulation method. Optimized formulations D-6 & D-10 showed very good stability profile as well.

Keywords: Dimenhydrinate, Gastro-retentive, Floating tablet, Direct compression, Melt granulation.
