Abstract

IDENTIFICATION AND SEPARATION OF QUERCETIN FROM ETHANOL EXTRACT OF CARMONA RETUSA BY THIN LAYER CHROMATOGRAPHY AND HIGH PERFORMANCE LIQUID CHROMATOGRAPHY WITH DIODE ARRAY DETECTION

ABSTRACT

High Performance Liquid Chromatography (HPLC) is a chromatographic technique used to separate the components in a mixture, to identify each component and to quantify each component in which the stationary phase consiststed of small particle (1.8 – 15 μm) packing contained in a column with a small bore (2 – 4.6 mm), one end of which is connected to liquid eluent (mobile phase). The analysis of Quercetin was conducted on Agilent 1100 HPLC system (Chemstation software) equipped with universal injector, injection volume 10 μl and DAD (Diode-Array Detector) - UV-VIS detector. HPLC method was developed for the quantitative estimation of Quercetin from alcoholic extract of Carmona retusa, with octadecylsilane (250 x 4.6 mm, C18 Inertsil ODS- 3V) as a stationary phase and mobile phase for Quercetin was used 0.05% orthophosphoric acid and methanol with gradient elution. Our quantitative HPLC analysis revealed that the Carmona retusa sample contained 995 ± 1.17 μg/g of quercetin.
