Abstract

IN-SILICO DESIGN AND SYNTHESIS OF NOVEL 2-HYDROXY QUINOXALINE BEARING THIAZOLIDINONE DERIVATIVES AS POTENTIAL ANTIDIABETIC AGENTS

Poornima T.*, Manju P. T. and Anjana E.

ABSTRACT

The present work aims to design and synthesize some novel quinoxaline bearing thiazolidinone derivatives and to perform in-vitro evaluation of their antidiabetic activity. In-silico design of derivatives was carried out using various software’s such as ACD lab Chemsketch, Molinspiration, PASS (Prediction of activity spectra for substances) and Auto Dock Vina. In the present study about 10 compounds were designed and from this, 5 derivatives possessed drug likeness properties, obeyed Lipinski rule of five and exhibited high docking score. These derivatives were selected for synthesis. The synthesized compounds were purified by recrystallization from ethanol and dimethyl formamide. Characterization of derivatives were done by determining its melting point, retention factor from thin layer chromatography and spectral studies (Infrared, Proton NMR spectroscopy, Mass spectroscopy). In-vitro antidiabetic evaluation of all the derivatives were done by PPARš¯›¾ ELISA study and all of them showed better activity when compared with standard.

Keywords: Quinoxaline, Thiazolidinone, Docking, AutoDock Vina, PPAR????.
