EFFECT OF GERMINATION ON SORGHUM LIPOXYGENASE (EC1.13.1.13) ACTIVITY

ABSTRACT
Lipoxygenase (EC1.13.1.13) are ubiquitous non heme iron containing dioxygenase that catalyse the hydroperoxidation of polyunsaturated fatty acids containing a cis, cis–1, 4 , pentandiene moiety. The crude lipoxygenase was extracted from 96h germinated sorghum and also from ungerminated sorghum in a phosphate buffer of pH 6.5. The extract was centrifuged at 2000rpm for 30 minutes .The purification of crude lipoxygenase extract was done by ammonium sulphate precipitation, dialysis, gel fitration on sephadex G-25. The enzyme recorded an optimum pH and temperature of 7.0 and 300C respectively. The activity was done on linoleic acid using spectrophotometer.
