ESTIMATION OF ANTI-INFLAMMATORY AGENT BY DERIVATIVE AND SIMPLE UV SPECTROSCOPIC METHODS

ABSTRACT
A newly synthesized anti-inflammatory agent (Methyl 2-(p-
Methoxylanilino)-5-(4-methoxybenzoyl)- 4-methylthiophene-3-
carboxylate) showed anti-inflammatory activity at a very low dose as
compared to both of the standard drugs mefenamic acid and
ibuprofen. So for further development of this drug simple, precise,
robust and accurate methods has been developed. The drug was
estimated by Simple and Derivative UV spectroscopy using Shimadzu
UV 1800 spectrophotometer using matched cuvette of 1 cm path
length. Simple UV Spectroscopic method is developed at 228nm and
Derivative Spectroscopy is developed at 360.6nm/398.6nm in
methanol. The method was validated as per ICH Guideline. The
linearity-range for the drug was found to be2-10 μg/ml for both the
methods. The correlation co-efficient for both simple and derivative
spectroscopy is 0.999 proving good linearity between concentration of drug and response for
both the methods. The methods are accurate within the range 2-10μg/ml. The % RSD values
of repeatability, intermediate precision and reproducibility were less than 2, which indicate
that both the methods has good reproducibility. Both the methods are robust and withstands
the small but deliberate changes. Thus the proposed methods are simple, accurate and precise
and suitable for routine analysis of the drug.
