QUALITY BY DESIGN IN PHARMACEUTICAL DEVELOPMENT: PRINCIPLES, TOOLS, REGULATORY FRAMEWORK, AND EMERGING APPLICATIONS
Ms. Samruddhi Pate*, Ms. Yogini Matale, Ms. Payal Ghare, Ms. Dhanshri A. Kalsai,
ABSTRACT Quality by Design (QbD) is a science- and risk-informed method of developing medications. It is an alternative approach to relying mainly on finished product testing by embedding quality into the formulation and manufacturing process of a product. We cover this review of pharmaceutical QbD core principles and steps of applying them, such as Quality Target Product Profile (QTPP), critical quality attributes (CQAs) identification, materials and process conditions assessment, risk assessment, and focused studies to understand the impact of these factors. Development and performance can be guided by tools and techniques, which include Design of Experiments (DoE) to identify the critical quality attributes (CQAs) of the product, failure mode and effects analysis, Process Analytical Technology (PAT), statistical process control, and multivariate analysis. FDA guidance and ICH guidelines—including Q8(R2), Q9(R1), Q10, Q12, Q13, Q14, and Q2(R2)—sustain product development, process control, and the route of alwaysimproving through a cycle. Though QbD can enhance understanding of the processes and lead to more consistent production, its application in practice demands substantial resources, quality data, teams, and regulatory navigation. Emerging technologies, including digital analytics, machine learning, digital twins, and continuous manufacturing, present additional opportunities. Called a well-validated models, good data governance, and o regulatory oversight commensurate with risk. Even so, QbD finds its best use when the science is established, quality risk management is applied, and continual performance monitoring is part of an effective quality system.
