ABSTRACT

Periodontitis is a chronic inflammatory disease characterized by the destruction of the supporting structures of teeth due to bacterial infection. Local drug delivery systems have gained importance as adjuncts to mechanical periodontal therapy because they provide high drug concentrations at the diseased site with minimal systemic side effects. The present formulation combines diclofenac, doxycycline, and metronidazole in a gel base to achieve anti-inflammatory, antibacterial, and antimicrobial effects. Diclofenac reduces inflammation and pain by inhibiting prostaglandin synthesis, while doxycycline provides broad-spectrum antibacterial activity and inhibits collagenase activity, thereby preventing tissue breakdown. Metronidazole is effective against anaerobic bacteria commonly associated with periodontal infections. The synergistic action of these agents in a gel formulation enhances periodontal healing, reduces pocket depth, and improves clinical outcomes. This combination of gel represents an effective adjunctive therapy in the management of periodontitis.
