A REVIEW ON COLON-SPECIFIC ORAL DRUG DELIVERY SYSTEM: DESIGN STRATEGIES AND EMERGING DEVELOPMENTS

ABSTRACT Colon-targeted drug delivery systems (CDDS) have received significant attention because of their capability in treating local diseases such as inflammatory bowel disease (IBD), colorectal cancer, and systemic conditions that advantage from colonic absorption. This overview explores diverse design strategies and emerging developments in oral drug delivery targeted to the colon. It begins with an outline of the colon’s anatomy and physiological environment, which plays a vital function in drug absorption and metabolism. The selection of polymers for colon targeting is essential, and various pH-sensitive, biodegradable, and time-dependent polymers are discussed. Several techniques are reviewed in detail, including pH-dependent systems, time- managed release, microbiota-triggered mechanisms, and pressure-controlled drug delivery systems. Recent improvements, together with the integration of hot-melt extrusion (HME) and 3-D printing technologies, offer novel solutions for precise and effective drug targeting. These emerging techniques enhance drug stability, bioavailability, and patient compliance. This review gives insights into the challenges and future perspectives of colon-targeted drug delivery, emphasizing the need for revolutionary strategies to optimize therapeutic efficacy.
