Abstract

VALIDATED HPTLC METHOD FOR SIMULTANEOUS DETERMINATION OF OFLOXACIN AND DEXAMETHASONE SODIUM PHOSPHATE IN EYE DROPS

ABSTRACT

A new, simple, sensitive, precise and accurate High-performance thin-layer chromatographic method for simultaneous determination of ofloxacin and Dexamethasone sodium phosphate in their combined dosage forms has been developed, validated and used for determination of the compounds in commercial pharmaceutical products. Chromatographic separation was performed on aluminium plates precoated with silica gel 60 F254 as the stationary phase and chloroform:acetonitrile:1-butanol:formic acid (3:1:3:3, v/v/v/v) as a mobile phase. Densitometric measurements of their spots were achieved at 270 nm over the concentration ranges of 1200-3000 ng spot-1 and 400-1000 ng spot-1, with mean recoveries of 98.93 ± 2.44 and 98.79 ± 2.79 for Ofloxacin and Dexamethasone sodium phosphate respectively. Limit of detection for Ofloxacin and Dexamethasone sodium phosphate were found to be 80.36 ng spot-1 and 101.76 ng spot-1 respectively.
