Abstract

DEVELOPMENT AND CHARACTERIZATION OF ANTIDIABETIC MOUTH DISSOLVING TABLET

ABSTRACT

Metformin hydrochloride is an orally administered antihyperglycemic agent, used in the management of non-insulindependant (type-2) diabetes mellitus. Difficulty in swallowing (dysphagia) is common among all age groups, especially in elderly and pediatrics. Unfortunately, a high percentage of patients suffering from type-2 diabetes are elderly people showing dysphagia. In this study, Mouth dissolving tablets were prepared using direct compression and wet granulation method. Methods of preparation along with superdisintegrants are the major players in the success of this dosage form. The tablets of Metformin hydrochloride were prepared using different superdisintegrants by direct compression and wet granulation.Ten formulations were prepared by using both techniques (Direct compression and wet granulation). Pre-compression studies including Bulk density, Tapped density, Angle of repose, Carr‟s index, Hausner‟s ratio and compatibility studies such as Fourier Transform Infrared spectroscopy (FTIR) were performed. Post compression studies on various parameters i.e. tablet hardness, weight variation, friability, disintegration time, dissolution studies wetting time, water absorption ratio, modified disintegration, uniformity of contents were conducted. Through these study we have known that batch P4 which is prepared by wet granulation, is the best formulation in all ten batches. In vitro drug release of best formulation (P4) was found to be 99.27% containing crospovidone and sodium starch glycolate as superdisntegrants in the formulation and drug release within 12 min. These research demonstrated effective use of Metformin hydrochloride mouth dissolving tablet and as an ideal drug release formulation for diabetes and it also represents potential of Mouth dissolving tablets in promoting rapid absorption, improved bioavailability, effective therapy on population and patient compliance at large. So from this work it was concluded that Mouth dissolving tablets of metformin hydrochloride were prepared successfully.
